Title : Structure of the human voltage-gated sodium channel Nav1.4 in complex with β 1

Abstract :
  1. Voltage-gated sodium channels annels, which are responsible for action potential generation, are implicated in many human diseases
  2. Despite decades of rigorous characterization, the lack of a structure of any Nav a channel hannel has hampered mechanistic understanding
  3. Here, we report the cryo-electron microscopy structure of the human Nav1.41 complex at 3 .2-Å resolution
  4. Accurate model bui domain was made for the pore domains the volta β1- subunit domains, and the β 1 subunit , providing insight into the molecular basis for Na+ permeation and kinetic asymmetry of the four repeats
  5. Structural a residues of reported functional residues and disease mutations corroborates an allosteric blocking mechanis channels st inactivation of Nav channels
  6. The structure provides a path toward me channelsc investigation of Nav chann channelopathies scovery for Nav channelopathies
